Well, let's start with this first:

My SLR experience is with Minoltas (35mm film) and now a Nikon D300.

I can't give you too many specifics about Canon DSLR's, because I've never owned one or shot with one (other than at a camera store.) I've got nothing against them, and almost bought a 50D recently. The layout of the dials on the Nikons is more similar to what I was used to with Minoltas, where the Canon was quite different with the back dial. That, combined with marginally better weather sealing on the Nikon D300 vs. the Canon 50D, and I decided to go with Nikon. We do get a touch of rain here in Seattle.

Also, I'm left-eye dominant but right handed. That means I'm looking through the viewfinder with my left eye, and squishing my nose against the back of the camera. I noticed on the 50D that my nose was right on the multi way controller. I still hit a control dial on the Nikon, but it is one I can lock out, and it was more comfortable on my schnaz. Maybe the Canon dial was able to be locked out too, but I'm not sure.

wouldn't the D300 be better compared with the 5D MkII since they are both full frame or would that be the D700?

I just don't like Canons because I started out with a Nikon. If my dad bought an XTi instead of a D60 that day, I'd be a Canon guy today...

But still, if you're talking beginner level DSLRs, Nikon wins over build and material quality, and Canon wins over lens compatibility.

Go for Canon -- from my experience, lenses are cheaper than Nikon.

That said, I have a Canon XSi that is great. It's my first DSLR, and is $650 with lens and free shipping from Amazon.com (who is an authorized Canon seller).

I'd say Nikon is king of the hill in lens compatibility - I can use 30 year old glass on my D200. The Nikon mount hasn't changed in....forever? I'm not sure. Certainly you give up some features, like matrix metering and autofocus in the older lenses, but they still work. There are entire generations of lenses that Canon has obsoleted.

50 years (1959). ;]

it's pretty cool to be able to get a little D40 for under $400 and a $50 used lens from the 60's and get amazing results. wish they made something smaller than the D40 (a la olympus e-420) to carry around in the glovebox or [jacket] pocket.

That's lens selection, not compatibility. But it probably outweighs what I mentioned about Canon.
